    IPR Pérola: Dwarf Arabica coffee cultivar with high resistance to leaf rust and large beans

    IPR Pérola is a Coffea arabica cultivar developed from a cross between IAPAR 59 and Mundo Novo IAC 376-4. It features a dwarf-medium size, high yield, excellent cup quality, a medium-early ripening cycle, and large beans. Additionally, it boasts high resistance to coffee leaf rust.

    New and efficient AS-PCR molecular marker for selection of coffee resistant to coffee leaf rust

    Coffee leaf rust, caused by Hemileia vastatrix, is the most important disease affecting Coffea arabica. The use of molecular markers through marker-assisted selection (MAS) is an effective approach to develop cultivars with multiple and durable resistance, which is economically and environmentally beneficial for this disease control. In this work, a new molecular marker based on allele-specific polymerase chain reaction (AS-PCR) associated with the quantitative trait locus (QTL) of the linkage group 5 (LG5), which confers resistance to races I, II and pathotype 001 of H. vastatrix, was developed. The CaRHv10-AS marker was validated in a segregating genetic mapping population of 247 F2 genotypes and demonstrated selection efficiency greater than 97% for genotypes with resistance to three races. Thus, this work provides a fast, robust, and affordable molecular marker for use in MAS, facilitating the development of coffee plants with multiple resistance and ensuring sustainable coffee production.

    New vegetation index for monitoring coffee rust using sentinel-2 multispectral imagery

    Coffee Rust (Hemileia vastatrix) is considered the primary coffee disease in the world. The pathogenic fungus can find favorable environmental conditions in different countries, constantly threatening coffee producers. The previous detection of the incidence of coffee rust in a region is crucial because it provides an overview of the disease’s progress aiding in coffee plantations management. The objective of this work was the development of a vegetation index for remote monitoring of coffee rust infestation. Using satellite images from the MSI/Sentinel-2 collection, the Machine Learning classifier algorithm - Random Forest, and the cloud processing platform - Google Earth Engine, the most sensitives bands in coffee rust detection were determined, namely B4 (Red), B7 (Red Edge 3) and B8A (Red Edge 4). Thus, the Triangular Vegetation Index method was used to create a new vegetative index for remote detection of coffee rust infestation on a regional scale, named Coffee Rust Detection Index (CRDI). A linear regression model was created to estimate rust infestation based on the performance of the new index. The model presented a coefficient of determination (R²) of 62.5%, and a root mean square error (RMSE) of 0.107. In addition, a comparison analysis of the new index with eight other vegetative indices commonly used in the literature was carried out. The CRDI obtained the best performance in coffee rust detection among the others. This study shows that the new index CRDI has the robustness and general capacity to be used in monitoring coffee rust infestation on a regional scale.

    Coffea arabica clones resistant to coffee leaf miner

    Brazilian arabica coffee production is based on a set of highly productive cultivars sexually propagated. All of them are susceptible to coffee leaf miner, Leucoptera coffeella, and most of them are also susceptible to coffee leaf rust, Hemileia vastatrix. Aiming to reduce the selection process of cultivars with multiple resistances to both biotic constraints, the Coffea arabica clones 760, 1059, 1064 and 1215 were evaluated under field conditions. The mean yield of four C. arabica clones was not statistically different from the experimental controls; however, the yield of clone 1059 was 21% higher than the control cultivar Obatã IAC 1669-20. Leaf miner resistance level in all the evaluated clones remained the same as the mother plant’s. However, the same did not occur for resistance to H. vastatrix, due to the emergence of new pathogen races. In summary, it is potentially feasible the adoption of clone 1059 as a new clonal cultivar resistant to coffee leaf miner.

    Assisted selection using molecular markers linked to rust resistance SH3 gene in Coffea arabica

    The aim of this work was to validate markers linked to the SH3 gene of coffee leaf rust (CLR) resistance and use them in assisted selection. Initially, we validated the markers in genotypes already known to carry SH3. Next, we performed phenotype and genotype evaluation for resistance to CLR in coffee plants growing under field conditions. We used Arabica coffee progenies derived from BA-10, which carries the SH3 gene due to introgression of C. liberica. Three SCAR markers (SP-M16-SH3, BA-48-21O-f, and BA-124-12K-f) and one SSR marker (Sat244) linked to SH3 gene were used to amplify the coffee plants’ DNA. Our assessments of markers validation in resistant genotypes, SP-M16-SH3 and BA-124-12K-f, were efficient to identify the SH3 gene. These two markers were used to evaluate the progenies derived from BA-10 and were significantly linked to the phenotype evaluations. The SP-M16-SH3 marker was more efficient, with the advantage of being codominant.

    Temporal progress of coffee leaf rust and environmental conditions affecting severity in Veracruz State, Mexico

    Coffee is an important crop in Mexico. Unfortunately, coffee production has been affected by coffee leaf rust (CLR). For Veracruz, the second state in Mexico with the major production of coffee, there are available reports of weekly CLR severity, but these are only informative without in-depth inferential analysis. We analyzed variations of CLR severity along the year in Veracruz with data from municipal weekly reports provided by Mexico’s federal government phytosanitary epidemiological monitoring coffee program. We selected reports dated in 2018 from nine municipalities and after calculations of mean monthly severity values, we conducted a one-way ANOVA (months as factors) of severity data. We compared this information with other coffee-producing regions. Additionally, we explored the association of temperature, rainfall, and altitude with CLR severity using Principal Component Analysis and multiple linear regressions. Temperature and rainfall data were obtained from Mexican National Meteorological Service. We found that CLR severity in October, November, December, and January (months of harvest period) was significantly higher than values from March-June. During the harvest period, coffee plants allocate resources mainly for fruiting which competes in resources for other tasks such as defense and leaf growth, so this competition of resources can explain the positive relationship found between fruit load and CLR severity. This monthly variation of severity was similar to those reported in Chiapas, Guatemala, Colombia, Uganda, and Ethiopia. Our model (R2 = 0.948) showed a significant and negative effect of minimum and maximum temperature (in a range from 9.9 – 15.5 °C and 18.5 – 26.5 °C, respectively) on CLR severity, while the effect of rainfall (in a range from 32.0 – 359.9 mm) and médium temperature (from 14.3 – 20.5 °C) was positive. With our study, we suggest applications of fungicides in March-June when coffee plantations are in leaf phenophase.

    Management of coffee leaf rust using L-glutamic acid biofertilizer combined with fungicide

    The use of biofertilizers is a potential tool for the management of crop diseases. Coffee leaf rust, which is commonly controlled by triazole and strobilurin fungicides, is one of the main phytosanitary challenges associated with coffee cultivation. However, the indiscriminate use of such fungicides may be harmful to the environment and human health, in addition to having a negative impact on coffee exports. The aim of this study was to evaluate the effect of foliar application of L-glutamic acid on the incidence and severity of coffee leaf rust in the southern region of Minas Gerais, Brazil. A biofertilizer made of sugarcane molasses fermented by the bacterium Corynebacterium glutamicumwas used in combination with 25% L-glutamic acid and a fungicide of the triazole group registered for crops. The experimental design adopted was randomized blocks with four replications (eight plants per replicate) and seven treatments: fungicides, control test, 0.8 L ha-1 of biofertilizer, combinations of 0.04, 0.06, 0.08, and 0.1 L ha-1 of biofertilizer and fungicide.Three treatments were initially applied in a preventive way, before the rainy season, and then at intervals of 60 days. After seven months of treatment, we observed that coffee leaf rust incidence was significantly lower in coffee plants treated with combined products when compared to treatments of either fungicide or biofertilizer only, with a reduction of 56% and 45%, respectively, being observed. Among the combinations of biofertilizer and fungicide, coffee leaf rust incidence reduced with an increase in the biofertilizer dose, such thatthe disease incidence in plants treated with a dose of 0.1 L ha-1was 58% lower than that in plants treated with fungicide only. No differences in disease severity were observed among the treatments. In conclusion, the use of a combination of biofertilizer and fungicide is more effective for the management of coffee leaf rust than the use of the isolated products.

    Avaliação de produtos alternativos no controle da ferrugem do cafeeiro

    Objetivou-se neste trabalho avaliar produtos orgânicos quanto ao efeito protetor e indutor de resistência à ferrugem do cafeeiro (Hemileia vastatrix), comparando-se com fungicidas sistêmicos. O trabalho foi desenvolvido em casa-de-vegetação, utilizando-se mudas de cafeeiro cv. Catuaí Vermelho 144. Foram avalidados como indutores de resistência extratos foliares, suspensões de conídios fúngicos e de células bacterianas, fertilizantes foliares, hipocloritos e acibenzolar-S-methyl (ASM). O delineamento experimental utilizado foi em blocos casualizados com três repetições, sendo que cada parcela experimental foi constituída de cinco mudas de café perfazendo um total de 15 mudas por tratamento. Os experimentos foram repetidos duas vezes com resultados semelhantes. Quando avaliado o efeito protetor, a maioria dos compostos reduziu o número de pústulas e a área foliar com ferrugem em comparação à testemunha 1 (água). Extratos de folhas de café, obtidos pelo processo aquoso e extratos de sementes de Azadirachta indica ('neem") obtidos pelo processo de extração com metanol apresentaram proteção similar, mas nunca superior a epoxiconazole + piraclostrobin. Extratos aquosos de folhas de café, ASM, Bacillus subtilis e Pseudomonas putida reduziram a infecção causada por H. vastatrix em mais de 77 %.

    Expansão da pústula da ferrugem em três cultivares do cafeeiro

    A expansão da lesão é considerada importante componente epidemiológico em vários patossistemas. Com o propósito de avaliar a expansão da pústula da ferrugem (Hemileia vastatrix) em três cultivares de cafeeiro e o valor das respectivas taxas de expansão da lesão (TEL) sob condições controladas delineou-se este estudo. O experimento foi instalado em câmaras de crescimento de plantas com temperatura de 20±2 ºC e umidade relativa de 70%. Foram inoculadas vinte plantas de cada uma das cultivares Acaiá, Catuaí e Rubi, de três meses de idade, com 0,5mg/mL da suspensão de urediniósporos. Após 42 dias da inoculação, sete pústulas foram marcadas aleatoriamente, em 7 plantas de cada cultivar e, em seguida, fotografadas na face abaxial da folha, com câmara digital, durante 15 dias, para posterior mensuração da área, utilizando o programa Image Tool da UTHSCA. Todas as imagens digitalizadas foram padronizadas em 4,0 x 4,0 cm. Ao final do experimento, foi avaliada a produção de urediniósporos/cm² de pústula, em hemacitômetro. As curvas de progresso da expansão da pústula nas três cultivares foram ajustadas ao modelo exponencial. A TEL foi de 0,14, 0,16 e 0,16 mm²/dia e o número de urediniósporos produzidos foi de 918,6, 929,3 e 934,8/cm² de pústula, para as cultivares Acaiá, Catuaí e Rubi, respectivamente. Não houve diferenças, entre as três cultivares, consideradas suscetíveis, em relação à TEL e à produção final de urediniósporos. Estes resultados podem contribuir para maior compreensão dos possíveis mecanismos que atuam no aumento da severidade da ferrugem do cafeeiro, sem que ocorram novas infecções.

    Resistência de progênies de café Catimor à ferrugem

    Visando avaliar o desempenho de genótipos de café Catimor, foram realizadas avaliações em nível de campo quanto à capacidade produtiva e a incidência da ferrugem e, em casa-de-vegetação os componentes da resistência quantitativa à raça II do patógeno. A reação à infecção foi avaliada por meio de uma escala de notas de 0 a 9, sendo 0 a 3 para plantas consideradas resistentes e 4 a 9 para suscetíveis. Os componentes da resistência quantitativa avaliados foram: freqüência de infecção, esporulação, área foliar lesionada, período de incubação e de latência. As progênies UFV 5550, UFV 6861, UFV 6870, UFV 6831 e UFV 6834 apresentaram produtividade semelhante à da cultivar Catuaí utilizada como testemunha (P < 0,05). Nenhuma planta das progênies UFV 5530, 5451, 5550, 6903 e 5464 apresentou pústulas com esporos. Os descendentes das progênies em estudo variaram quanto à reação à ferrugem, desde resistência completa a alta suscetibilidade, sendo que a maioria situou-se nos níveis intermediários, evidenciando a presença de resistência quantitativa. O número médio de lesões e a área foliar lesionada foram 13 e 21 vezes menores, respectivamente, em UFV 6866. A progênie UFV 6870 apresentou a nota 0,1 de esporulação e a cv. Catuaí, 3,6. O período de incubação nos genótipos estudados variou de 18 a 36 dias e o período latente variou de 20 a 46 dias. Comparado à cultivar Catuaí Vermelho IAC 15, conclui-se que a maioria dos genótipos de Catimor estudados possuem resistência quantitativa.
